Product Range

CST provides primary and secondary antibodies and their conjugated forms, and many types of kits, for example, PathScan ELISA kits, based on their high-quality, highly validated primary antibodies. The kits include those for detecting endogenous levels of the members of various signaling pathways, cytokines and growth factors, and various assay kits for protein activities, immunohistochemical imaging, chromatin IP, cell viability, proliferation and apoptosis, and other cellular activities, as well as siRNA for selected gene targets. It also provided (now discontinued) PathScan antibody array kits such as PathScan RTK Signaling Antibody Array Kit and PathScan Akt Signaling Antibody Array Kit [1], which enable simultaneous detection of multiple proteins involved in specific pathways [1].

Company Information and History

Cell Signaling Technology, Inc. (CST) was founded in 1999 by scientists in the Cell Signaling group at New England Biolabs (NEB). CST's founder is Dr. Michael Comb. New England Biolabs was established in 1974 by and owned by Dr. Donald G. Comb. Nobel Laureate Dr. Richard Roberts joined NEB in 1992 and is the Chief Scientific Officer of NEB.

The executive management of CST includes Dr. Michael J. Comb President / Chief Executive Officer, Mr. Fenel Eloi Chief Operating Officer / Chief Financial Officer, and Dr. Roberto Polakiewicz Chief Scientific Officer.

Unlike most reagent companies, CST and its parent company, NEB, maintain rigorous research programs, similar to a research institute or an academic department. Its scientists have published in recent years in Nature Cell Biology [2], Elife [3] among others.

CST develops and maintains PhosphositePlus PTM database (http://www.phosphosite.org/) [4], an online database of protein post-translational modifications (not just protein phosphorylation). NIH grants partially funded the database. It catalogs the protein post-translational modifications from the literature and also from high-throughput screening experiments. It is the most comprehensive and most accessed protein modification database.

Cell Signaling Technology products have been widely used in research and diagnostic laboratories. For example, R Divine et al utilized the Cell Signaling anti-phospho-Akt (S473) (D9E) XP rabbit monoclonal antibody ( 4060) with a ProteinSimple Jess instrument [5]. van de Kooij B et al detected the tyrosine phosphorylation of Ned10 with Cell Signaling Technology rabbit monoclonal anti-phosphotyrosine antibody (Cat 8954) at 1:2000 dilution [6]. Lee YR et al stained tumor paraffin sections with anti-pAKT antibody ( 4060) and anti-cleaved caspase 3 ( 9661) antibodies from CST to study the reactivation of PTEN tumor suppressor [7].

Among those citing Cell Signaling Technology reagents, the top 5 most cited antibodies, without any doubt, are involved in cellular signaling pathways (Table 2). They are AKT antibodies, ERK2 antibodies, ERK1 antibodies, and JNK antibodies. These numbers of publications are those that can be identified by Labome. The actual numbers for each product are much higher. Some of recent examples are listed here. L Zhao et al applied PR A/B ( 3176), GR ( 3660), AKT ( 4691, 4060), P21 ( 2947), β-catenin ( 8480), FoxA1 ( 53528), HER3 ( 4791, 12708), HER2 ( 2247, 4290), caspases ( 9661, 8438, 9496, 9505), cleaved PARP ( 5625) and GAPDH ( 8884) antibodies in western blotting [52]. Kitchen P et al used Cell Signaling anti-calmodulin antibody D1F7J, anti-lamin B1 antibody D4Q4Z and anti-alpha tubulin antibody (2144) for their study of AQP4 in CNS edema [53]. Rhodes JDP et al used its rabbit mAb anti-RING1B antibody ( 5694) for ChIP at 1:1000 and mouse mAb anti-H3S10p antibody (9706) for flow cytometry at 1:25 [54]. Moro A et al used Cell Signaling Technology rabbit monoclonal Ago2 antibody clone C34C6 ( 2897) to label AGO2 in HUVEC lysates and cells in Western blots and immunocytochemistry [55]. Its GAPDH antibodies, used as loading controls, gain popularity rapidly in recent years [55-60]. Anti-HSP90 antibody ( 4874) is also used as a loading control.
