This is a knockout-validated antibody summary, based on the publication "Intercellular transfer of pathogenic α-synuclein by extracellular vesicles is induced by the lipid peroxidation product 4-hydroxynonenal", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).
Mouse monoclonal IgG1, kapp
Company: Thermo Fisher
Antibody: α-synuclein
Catalog number: 32-8100
Summary: Mouse monoclonal IgG1, kappa against recombinant human alpha-synuclein. Reacts with human, mouse, non-human primate, and rat. Suitable for western blot, immunohistochemistry and ELISA.
Western blot
Control HEK cells (−), HEK cells overexpressing wildtype human α-synuclein (+), and α-synuclein knockout HEK cells (KO). Cells were lysed in radioimmunoprecipitation assay buffer (50 mM Tris, pH 7.4, 150 mM NaCl, 0.1% SDS, 1% NP-40, 2 mM EDTA, 1 mM DTT, 1 mM PMSF, 200 M Na3VO4, 50 mM NaF and protease inhibitors (Roche) on ice.
1:1,000 dilution overnight at 4°C.
HRP-conjugated secondary antibodies (Cell Signaling Technology).
Enhanced chemiluminescence kit (Thermo Fisher Scientific).
EVs were isolated from the medium of cultured control HEK cells (−), HEK cells overexpressing wildtype human α-synuclein (+), and α-synuclein knockout HEK cells (KO). Proteins in the EVs and the cells that released the EVs were separated in a NuPAGE Bis-Tris gel and immunoblotted using antibodies against α-synuclein, flotillin-1, calnexin and actin. Please see Figure 6a in the article [1].
The same clone (Syn 211) is sold as LifeSpan Biosciences LS-C26941; Abnova MAB1830; Invitrogen MA1-12874, AHB0261, 32-8100.
