This is a knockout-validated antibody summary, based on the publication "Deletion of the tyrosine phosphatase Shp2 in Sertoli cells causes infertility in mice", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

shp2 antibody | knockout validation | Santa Cruz Biotechnology sc-280 figure 1
Figure 1. (A) Shp2 expression was detected with immunofluorescence staining in the seminiferous tubules at embryonic day 18 (E18). Arrowheads indicated Sertoli cells. The cell nucleus was stained with DAPI. (B) Shp2 protein levels were analyzed by Western blotting in primary Sertoli cells and germ cells isolated from 14-day-old mice. Wt1 is a marker of SCs and SCP3 is a marker protein of GCs. Tubulin was used as a loading control. From [1].
Antibody information

Rabbit polyclonal IgG

Company: Santa Cruz Biotechnology

Antibody: shp2

Catalog number: sc-280

Summary: Rabbit polyclonal IgG against an epitope mapping at the C-terminus of SH-PTP2 of human origin. Recommended for detection of SH-PTP2 of mouse, rat, human and avian origin by western blot, immunoprecipitation, immunofluorescence, immunohistochemistry (paraffin) and ELISA. Also reactive with additional species, including and equine, canine, bovine, and porcine.

Validation Method

Western blot | Immunohistochemistry

Sample

Primary Sertoli cells and germ cells isolated from 14-day-old mice. For immunohistochemistry, paraffin-embedded testes were sectioned serially at 5 μm thick.

Blocking agent

Bovine serum albumin solution.

Primary incubation

WB: overnight at 4 °C.

IHC: overnight at 4 °C.

Secondary incubation

WB: horseradish peroxidase-conjugated secondary antibodies (Aves) at 37 °C for 2 hours.

IHC: Alexa488-conjugated secondary antibodies (Molecular Probes).

Detection

WB: visualized with enhanced chemiluminescence.

Disclaimer

If the antibody described in this summary is a polyclonal antibody, since polyclonal antibodies are of limited quantity, please inquire the supplier whether any current polyclonal antibody with the same catalog number is exactly the same as the one described in this summary. Sometimes, different bleeds or different animals are used, usually with a different lot number. In such cases, the result in this summary may not apply to the new antibody with the same catalog number.
