Western blot
WT and macroH2A1.2 knockout (KO) K562 cells. Cells were resuspended in 0.5 mL lysis buffer containing 10 mM HEPES (pH 7.4), 10 mM KCl, 0.5% NP-40 with phosphatase inhibitors (Calbiochem) and after separation of the nucleoplasmic fraction, chromatin-bound proteins were released from the DNA by addition of 0.2 N HCl.
