dsg2 antibody | knockout validation | Santa Cruz sc-53486

This is a knockout-validated antibody summary, based on the publication "Central role for GSK3β in the pathogenesis of arrhythmogenic cardiomyopathy", as cited below [1], and "Desmoglein-2 harnesses a PDZ-GEF2/Rap1 signaling axis to control cell spreading and focal adhesions independent of cell-cell adhesion" for western blot knockout validation (figure 1b) [2]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Antibody information

Mouse monoclonal IgG1 (kappa light chain)

Company: Santa Cruz

Antibody: dsg2

Catalog number: sc-53486

Summary: Mouse monoclonal IgG1 (kappa light chain) against A-431 cell shavings recommended for detection of desmoglein 2 of human origin by western blot, immunoprecipitation and immunofluorescence. Non cross-reactive with Desmoglein 1 or 3.

Validation Method

Immunohistochemistry

Sample

Formalin-fixed, paraffin-embedded myocardium.

Blocking agent

0.5% Triton-X, 3% hydrogen peroxide, and 5% bovine serum albumin, respectively, for 1 hour at room temperature.

Primary incubation

Overnight at 4°C.

Detection

Laser scanning microscopy (Zeiss LSM 510 Meta).

Figure

Immunohistochemistry of desmoglein-2 (DSG2) in myocardium showed a complete absence of protein and junctional distribution of DSG2 in Dsg2mut/mut mice compared with WT mice. Scale bar: 10 μm. Please see Figure 2a in the article [1].

Notes

Antibody was only used in IHC. Western blot was done with goat polyclonal anti-DSG2 (V-19, Santa Cruz, sc-14860). See details in the article.
