Soleus muscle from WT, hetero- and homozygous R349P (c.1045_1047delAGG>insCCC) desmin knock-in mice. Small amounts of snap-frozen tissue were pulverized in a mortar in the presence of liquid nitrogen, taken up in lysis buffer (5 mM Tris-HCl pH 6.8, 10 % SDS, 0.2 M DTT, 1 mM EDTA) and heated at 95 °C for 5 min.

Primary incubation

1:1,000 in TBS-T.

Notes

Is not exactly a KO, but a knock-in (R349P mutation). The mutation results in a marked decreased expression of the protein in the homozygous mice, which it is a KO de facto.
