Validation Method

Immunohistochemistry

Sample

Soleus muscles were dissected and fixed for 15 min in 2 % PFA in PBS and single muscle fibers were prepared.

Blocking agent

Mouse on Mouse (M.O.M.) Basic Kit (Vector Laboratories) according to the manufacturer’s instructions.

Primary incubation

1:100 diluted in “M.O.M. Diluent” for 1 h.

Secondary incubation

Goat anti-rabbit Alexa Fluor 488 (Life Technologies) for 1 h at room temperature.

Detection

LSM780 fluorescence laser scanning microscope (Carl Zeiss, Jena, Germany) equipped with a Plan-Apochromat 63 × 1.4NA objective lens. Images were obtained using the LSM780 module and the Zeiss ZEN software.

Notes

This case is not exactly a KO, but a knock-in (R349P mutation). The mutation results in a marked decreased expression of the protein in the homozygous mice, which it is a KO de facto.
