Validation Method

Western blot | Immunofluorescence

Sample

ActbL/L and ActbL/L Cre MEFs. For immunofluorescence, cells were fixed with cold methanol at −20°C for 10 min.

Blocking agent

IF: 3% BSA for 1 h.

Secondary incubation

WB: Fluorescently labeled secondary antibodies.

Detection

WB: Odyssey Infrared Imaging System (LI-COR Biosciences, Lincoln, NE).

IF: 20× (NA 0.75), 40× oil (NA 1.35), or 100× oil (NA 1.40) objective on a Delta Vision personalDV microscope using softWoRx 3.7.1 software (Applied Precision).

Notes

FITC-conjugated antibody AC15; Abcam (same clone) was used in KO validation by IF.
