Validation Method

Western blot

Sample

Skeletal muscle from WT and αB-crystallin/HspB2-KO mice. Tissues were lysed with 250 μl of RIPA lysis buffer including protease (Thermo Scientific #87786) and phosphatase inhibitor cocktails (Thermo Scientific #78420).

Primary incubation

1:1,000 dilution.

Secondary incubation

1:7,000 dilution anti-mouse IgG HRP conjugate (Promega; W402B).

Detection

Amersham ECL Western Blotting Detection Reagents (GE Healthcare; RPN2106) was used to detect chemiluminescence with HyBlot CL autoradiography film (Denville Scientific Inc.).
