WT and XIAP KO Ls174t cell lines. Cells were lysed in buffer (1% Triton X-100, 50 mM Tris-HCl pH 7.4, 150 mM NaCl, 12.5 mM β-glycerophosphate, 1.5 mM MgCl2, 10 mM NaF, 2 mM DTT, 2 mM sodium orthovanadate, 2 mM Ethylene glycol-bis (2-aminoethylether)-N,N,N′,N′-tetraacetic acid (EGTA), and Protease Inhibitor Cocktail (Roche)).

Blocking agent

5% nonfat milk for 1 h at room temperature.

Primary incubation

1:1,000 dilution at 4 °C overnight.

Secondary incubation

HRP-conjugated secondary antibody at room temperature for 1 h.

Detection

Enhanced Chemiluminescence.
