Western blot.

Sample

Platelets from wild type or VASP-null (VASP KO) mice. Platelets were lysed with an ice cold buffer composed of 50 mM Tris-HCl (pH 7.4), 1 % Triton X- 100, 100 mM NaCl, 10 mM MgCl2, 20 % glycerol, 1 mM Na3VO4, and 1 mM pefabloc, and Rap1b was immunoprecipitated.

Blocking agent

Phosphate-buffered saline supplemented with 0.2 % Tween 20 and 3 % milk.

Primary incubation

Overnight at 4 °C.

Secondary incubation

1:10,000 dilution peroxidase-conjugated goat anti-mouse IgGs for 1 h.

Detection

Enhanced chemiluminescence.
