Sample

B and T lymphocytes purified from the spleen of mice having Usp9X specifically deleted in B cells (using CD19.cre) or in all hematopoietic cells (using vavCre transgene). Cells were lysed in RIPA lysis buffer (50 mM Tris, pH 7.4, 150 mM NaCl, 2 mM EDTA, 1% NP-40, and 0.1% SDS) supplemented with protease (Roche) and phosphatase inhibitors (Roche).

Secondary incubation

1:2,500 dilution of HRP-conjugated anti–rabbit IgG secondary (Cell Signaling Technology) or HRP-conjugated anti–rat IgG secondary (Santa Cruz Biotechnology, Inc.)
