Sample

WT and Thbs4−/− bone-marrow-derived macrophages (BMDM). Cells were harvested and lysed on ice using RIPA lysis and extraction buffer (# 89901, Thermo Scientific) supplemented with Halt™ protease and phosphatase inhibitor cocktail (#78442, Thermo Scientific).

Blocking agent

5% skim milk in TBST for an hour at room temperature.

Primary incubation

1:100 dilution overnight at 4 °C.

Secondary incubation

HRP anti-mouse secondary antibodies (#sc-516102, Santa Cruz Biotechnology).

Detection

Pierce® ECL western blotting substrate on X-ray films.
