Tau5 antibody | knockout validation | Biosource AHB0042

This is a knockout-validated antibody summary, based on the publication "Loss of Tau protein affects the structure, transcription and repair of neuronal pericentromeric heterochromatin" [1] (discussed below) and "A validated antibody panel for the characterization of tau post-translational modifications" [291572770 (see table 1 in the article for a summary of various Tau antibodies). Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Tau5 antibody | knockout validation | Biosource AHB0042 figure 1
Figure 1. Single confocal sections of nuclei from primary culture of KOTau neurons either non-infected (KO) or infected with lentiviral vectors encoding for hTau protein (KOhTau) or hTau protein fused to a nuclear localization signal (NLS) (KOhTauNLS) indicate that the clustered distribution of H3K9me3 in KOTau neurons is rescued in the presence of a nuclear localization of hTau protein. Tau5 antibodies were used to visualize total Tau protein and GFP fluorescence was used to visualize KOTau neurons. From [1].
Antibody information

Mouse monoclonal IgG1

Company: Biosource

Antibody: Tau

Catalog number: AHB0042

Summary: Mouse monoclonal IgG1 against purified bovine microtubule-associated proteins. Reacts with bovine, human, mouse, sheep, and rat. Suitable for western blot, immunoprecipitation and immunohistochemistry (paraffin). Published to react also with primates and to be suitable also for immunofluorescence and ELISA.

Validation Method

Immunocytochemistry

Sample

Wild-type and knock-out Tau murine primary cortical neurons. Cells were fixed with 4% paraformaldehyde.

Secondary incubation

Alexa 555-conjugated donkey anti-mouse (A31570, Molecular Probes).

Detection

Confocal laser scanning microscopy using an AxioImager Z2 (Zeiss LSM710 confocal system) equipped with a 63× lens, 1.4-numerical-aperture oil immersion lens (Plan Neofluor).
