Heart and kidney from tamoxifen-treated and -untreated TRPV2flox/flox or TRPV2flox/flox;MerCreMer+/− (TRPV2flox/flox; Cre+/−) mice. For WB: homogenized in a Hiscotron homogenizer (NITI-ON) in lysis buffer containing 20 mM HEPES (pH 7.4), 150 mM NaCl, 1% sodium deoxycholate, 1% SDS, 2 μg ml−1 leupeptin, 1 μg ml−1 aprotinin, 200 μM phenylmethylsulfonyl fluoride, and 200 μM benzamidine hydrochloride. For IHC: 5 μm frozen heart sections embedded in OCT compound (Tissue-Tek) were permeabilized with 0.1% Triton X-100 and incubated with primary antibodies.
1:200 dilution.
WB: chemiluminescence detection system (Perkin Elmer) and an LAS3000 Luminescent Image Analyzer (Fuji Film).
IHC: confocal microscope (Fluoview FV1000, Olympus) mounted on an Olympus IX81 epifluorescence microscope with a UPlanSApo × 60/1.35 oil immersion objective lens (Olympus).
