Immunohistochemistry - frozen section

The following information is for immunohistochemical staining.

Sample

Immunoblots of dorsal root ganglia (DRG) lysates and immunostaining of DRG sections prepared from WT and TRPV1-KO mice. L5 DRG from WT or KO mice were immediately post-fixed in 4% formaldehyde for 60 min at 4 C, then transferred to 30% sucrose overnight at 4째C. DRG were embedded in Tissue-Tek OCT compound and sectioned at 10 um by use of a Leica cryostat. The slices were stored at -20 C and further fixed in 4% formaldehyde for 10 min right before immunostaining.

Blocking agent

blocked for 2 h at room temperature in PBS containing 1% BSA, 5% horse serum and 0.3% Triton X-100

Primary incubation

overnight at 4 C

Secondary incubation

AlexaFluor-conjugated secondary antibodies or DyLight 594-conjugated IB4 in PBS containing 0.1 mM CaCl2, MgCl2, and MnCl2 at room temperature for 30 min

Detection

Images were acquired and quantified under a LSM 700 confocal microscope with a 20X objective lens (Carl Zeiss)
