Validation Method

Immunocytochemistry

Sample

Control and TREK-1 KO cultured astrocytes.

Blocking agent

0.1% Triton X-100 and 3% BSA for 1 h.

Primary incubation

1:100 dilution at 4 °C overnight.

Secondary incubation

1:400 dilution Alexa Fluor 594-conjugated secondary antibodies (Jackson Immunoresearch, West Grove, PA, USA) for 1 h at room temperature.

Detection

Confocal microscopy using a Nikon A1 confocal microscope (Nikon Instruments Inc., Melville, NY, USA).
