TRB-2 antibody | knockout examination | Santa Cruz sc-100878

This is a knockout-examined antibody summary, based on the publication "A Trib2-p38 axis controls myeloid leukaemia cell cycle and stress response signalling", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Antibody information

Mouse monoclonal IgG2b (lambda light chain)

Company: Santa Cruz

Antibody: TRB-2

Catalog number: sc-100878

Summary: Mouse monoclonal IgG2b (lambda light chain) against recombinant human TRB-2. Recommended for detection of TRB-2 of mouse, rat and human. Suitable for western blot, immunoprecipitation, immunofluorescence, immunohistochemistry (paraffin) and ELISA.

Note

KO validation is not ideal. Protein levels in the wt are not visible, and protein is only detected after overexpression.

Validation Method

Western blot

Sample

Whole cell lysates were prepared using ice-cold modified RIPA buffer (50 mM Tris, pH 8.0, containing 0.5% NP-40, 0.25% sodium deoxycholate, 150 mM NaCl, 1 mM EDTA, with protease and phosphatase inhibitors).

Secondary incubation

Anti-mouse IgG HRP-linked (GE-Healthcare, NXA931).

Detection

SuperSignal™ West Pico or Femto Chemiluminescent Substrates (Thermo Fisher Scientific) were used for signal detection on CL-XPosure™ radiography films. The films were developed using the Medical film processor SRX-101A (Konika Minolta, Tokyo, Japan).
