Sample

WB: Tissue protein samples were prepared in ice-cold lysis buffer (100 mmol/L TrisHCl pH7.4, 10 mmol/L NaCl, 1 mmol/L EDTA, 1 mmol/L EGTA, 10% glycerol, 0.5% deoxycholate, 1% Triton X-100, 0.1% SDS, 20mmol/L Na4P2O7, 2 mmol/L Na3VO4, 1 mmol/L NaF, 1 mmol/L PMSF) supplemented with 1X protease inhibitor and phosphatase inhibitor (Roche). IHC: antigen retrieval in tissue sections in boiling 10mmol/L sodium citrate (pH 6.0) for 30min.

Blocking agent

WB: 5% non-fat milk-TBS (w/v). IHC: 10% goat serum.

Primary incubation

WB: 1:2,000 dilution. IHC: 1:100 dilution.

Secondary incubation

WB: HRP-conjugated secondary antibodies. IHC: 1:200 dilution Alexa Fluor conjugated anti-mouse IgG (Invitrogen).

Detection

WB: enhanced chemiluminescence substrate (Thermo Sci.). IHC: DeltaVision (Deconvolution) image restoration microscope or a Zeiss Axioplan-2 imaging system.
