Figure 1. Immunoblot of NRF1 in WT and Nrf1-KO MEFs. In response to the proteasome inhibitor MG132, NRF1 protein significantly accumulates within cells. Two isoforms of NRF1 are present in MEFs, a high molecular weight form, and a cleaved low molecular weight nuclear form. Tubulin was used as loading control. Courtesy of the authors. The first 3 lanes are WT MEFs, the last 3 lanes are Nrf1 KO MEFs. Lanes 1 and 4 were treated with DMSO for 4 hours. Lanes 2, 3, 5 and 6 were treated with 10 uM MG132 for 4 hours. (TCF11 is the name of the human homologue of mouse Nrf1). The upper band is the non-cleaved cytoplasmic form of Nrf1. The lower band is the cleaved nuclear form.
