Sample

TCAB1 and Coilin in clonal KO HCT116 cells (with LhTRmin + TERT overexpression).

Blocking agent

WB: 4% nonfat milk (Carnation) in TBS buffer (150 mM NaCl, 50 mM Tris pH 7.5).

ICC: 4% BSA in PBS for 1 hr at room temperature or overnight at 4°C.

Primary incubation

WB: 1:2,000 dilution in blocking buffer at 4°C overnight.

ICC: 1:300 in blocking buffer for 1 hr at room temperature or overnight at 4°C.

Secondary incubation

WB: 1:10,000 diluted goat anti-rabbit IR Dye 800 (Rockland Immunochemicals, Pottstown, PA) in blocking buffer buffer for 1 hr at room temperature.

ICC: Alexa Fluor 568 (Thermo) secondary antibodies diluted in blocking buffer for 1 hr at room temperature.

Detection

WB: blots were scanned using a LI-COR Odyssey imager.

ICC: images were acquired using a Zeiss LSM510 Meta confocal microscope with a ×100/1.49 Apo objective, with 364-, 488-, and/or 543-nm laser excitation.
