Validation Method

Immunohistochemistry

Sample

Heart sections of of control and Tbx18cre/+;Tbx2fl/fl;R26mTmG/+ mice embryos. Embryos were fixed in 4% paraformaldehyde overnight and paraffin embedded for sectioning.

Primary incubation

1:1,000 dilution. Primary antibody was amplified using the Tyramide Signal Amplification (TSA) system from Perkin-Elmer (NEL702001KT, Perkin Elmer).

Secondary incubation

1:200 dilution Alexa488-conjugated anti-rabbit IgG (Invitrogen).

Detection

Leica DM5000 microscope with Leica DFC300FX digital camera.
