T-bet antibody | knockout validation | Santa Cruz Biotechnology sc-21749

This is a knockout-validated antibody summary, based on the publication "NK cells regulate CXCR2+ neutrophil recruitment during acute lung injury", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Antibody information

Mouse monoclonal IgG1 (kappa light chain)

Company: Santa Cruz Biotechnology

Antibody: T-bet

Catalog number: sc-21749

Summary: Mouse monoclonal IgG1 (kappa light chain) against full length recombinant murine T-bet. Recommended for detection of T-bet of mouse, rat and human origin by western blot, immunoprecipitation, immunofluorescence, flow cytometry and ELISA.

Validation Method

Western blot

Sample

Lung tissue from wild-type and Tbet-/- mice. Tissues were disrupted in T-Per Tissue Protein Extraction Reagent with Pierce Phosphatase and Protease Inhibitor (Thermo Fisher Scientific, Rockford, IL, USA).

Blocking agent

5% blotting-grade nonfat milk (Bio-Rad) in TBST for 1 h at room temperature.

Primary incubation

1:100 dilution overnight at 4°C.

Secondary incubation

1:10,000 dilution goat anti-mouse secondary antibody (31172; Thermo Fisher Scientific) for 1 h at room temperature.

Detection

Enhanced chemiluminescence (Thermo Fisher Scientific).
