Stat3α antibody | knockout validation | Cell Signaling 8768

This is a knockout-validated antibody summary, based on the publications "Ablation of STAT3 in Purkinje cells reorganizes cerebellar synaptic plasticity in long-term fear memory network", as cited below [1], and "Novel role of STAT3 in microglia-dependent neuroinflammation after experimental subarachnoid haemorrhage" for western blot knockout validation (figure 1c) [2]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Antibody information

Rabbit monoclonal IgG

Company: Cell Signaling

Antibody: Stat3α

Catalog number: 8768

Summary: Rabbit monoclonal antibody against a synthetic peptide corresponding to residues near the carboxy terminus of human Stat3α protein. Reacts with human, mouse, rat, hamster, and monkey. It is predicted to react based on 100% sequence homology with pig. Suitable for western blot, immunoprecipitation (IP), immunohistochemistry (paraffin), immunofluorescence/immunocytochemistry and chromatin IP.

Validation Method

Immunohistochemistry

Sample

WT and STAT3 Purkinje cell-KO mice cerebellum.

Blocking agent

5% normal goat serum (Vector Laboratories, Burlingame, CA) in PBS.

Secondary incubation

Biotinylated goat anti-rabbit IgG (BA-1000, Vector Laboratories, CA, USA).

Detection

Vectastain ABC kit (PK-4001, Vector Laboratories, CA, USA), DAB reagents (K5007, Dako, CA, USA), and counterstained with hematoxylin (S3309, Dako, CA, USA). Images were obtained using the Leopard program on a microscope (BX53; Olympus, Tokyo, Japan).
