Sample

Wildtype and SLFN11-knockout in DU145, CCRF-CEM, MOLT4 and EW8 cells. WB: whole cell lysates were prepared with the CelLyticâ„¢M lysis reagent (C2978, Sigma-Aldrich). IC: cells were fixed for 10 min with freshly prepared 4% (wt/vol) paraformaldehyde in phosphate buffered saline (PBS), and then permeabilized for 10 min in 0.5% Triton-X100/PBS.

Blocking agent

IC: 5% bovine serum albumin/0.1% Tween 20/PBS for 1 hour.

Primary incubation

IC: for 2 hours.

Secondary incubation

WB: horseradish peroxidase (HRP)-conjugated antibodies to mouse IgG (GE Healthcare, UK).

IC: Alexa Fluor 488 and/or Alexa Fluor 568 for 1 hour.

Detection

IC: confocal microscope (Nikon PCM2000).
