This is a knockout-validated antibody summary, based on the publications "GATA4 and GATA6 control mouse pancreas organogenesis", as cited below [1], and "Epigenetic modifier balances Mapk and Wnt signalling in differentiation of goblet and Paneth cells" for immunohistochemistry knockout validation (figure 6c) [2]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).
Company: Santa Cruz
Antibody: GATA4
Catalog number: sc-25310
Summary: Mouse monoclonal IgG2a raised against amino acids 328-439 of GATA-4 of human origin. Recommended for detection of GATA-4 of mouse, rat, and human origin by western blot, immunoprecipitation, immunofluorescence, immunohistochemistry (paraffin) and ELISA.

IHC-P, immunohistochemistry, paraffin sections.
Whole embryos or dissected pancreata collected at embryonic or adult stages were fixed in 4% paraformaldehyde in PBS overnight at 4°C, dehydrated with ethanol and xylene, and embedded in paraffin. Sections were cut at a thickness of 6 μm with a Leica DM6000B microtome.
Sections were dewaxed through a series of xylene and ethanol washes.
Dewaxed sections were boiled in antigen retrieval and incubated with 3% H2O2 for 15 min prior to block with 3% normal goat serum.
1:100, overnight at 4 °C in a humid chamber
Sections were washed three times with PBS and incubated with appropriate secondary antibodies at 1:300 dilutions in 3% normal goat serum at room temperature for 1 h.
Immunoperoxidase staining was performed using the Vectastain Elite ABC kit (Vector Laboratories) and developed using the peroxidase substrate DAB (Vector Laboratories).
