Validation Method

IHC-P, immunohistochemistry, paraffin sections.

Sample

Whole embryos or dissected pancreata collected at embryonic or adult stages were fixed in 4% paraformaldehyde in PBS overnight at 4°C, dehydrated with ethanol and xylene, and embedded in paraffin. Sections were cut at a thickness of 6 μm with a Leica DM6000B microtome.

Sections were dewaxed through a series of xylene and ethanol washes.

Blocking agent

Dewaxed sections were boiled in antigen retrieval and incubated with 3% H2O2 for 15 min prior to block with 3% normal goat serum.

Primary incubation

1:100, overnight at 4 °C in a humid chamber

Secondary incubation

Sections were washed three times with PBS and incubated with appropriate secondary antibodies at 1:300 dilutions in 3% normal goat serum at room temperature for 1 h.

Detection

Immunoperoxidase staining was performed using the Vectastain Elite ABC kit (Vector Laboratories) and developed using the peroxidase substrate DAB (Vector Laboratories).
