Validation Method

WB, western blot

Sample

FANCI-null HCT116 cells were lysed in 0.05 M Tris-HCl (pH 6.8), 2% SDS, 6% β-mercaptoethanol and boiled for 5min.

Blocking agent

blocking buffer (5% milk in TBS-Tween 20)

Primary incubation

Primary antibodies were diluted in blocking buffer (5% milk in TBS-Tween 20) and incubated overnight.

Secondary incubation

Horseradish peroxidase—conjugated anti-mouse.

Detection

Images were acquired using ImageQuant LAS4000 system (GE Healthcare)

Notes

FANCI-null HCT116 cells were generated through recombinant adeno-associated virus-mediated gene targeting.
