Western blot

Sample

293-TREx cells. Lysed in 10 µl of 1× Passive Lysis Buffer (Promega’s Dual-Luciferase Reporter Assay System, Cat: E1910).

Blocking agent

5 % milk in TBST, 1 h at room temperature

Primary incubation

1:1000–1:5000 dilution, in 5 % milk in TBST for 1 h, with 3 × 5 min TBST washes in between incubation steps.

Secondary incubation

Anti-Mouse-HRP-conjugated secondary antibody was from Cell Signaling (anti-mouse, Cat: 7076).

Detection

(ECL) Biorad ChemiDoc MP imager

Result

Clone 5.B5 selected and sequenced. HuR deletion confirmed.
