This is a knockout-validated antibody summary, based on the publication "Cellular response to alkylating agent MNNG is impaired in STAT1-deficients cells", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Figure 1. Western blot of Y701-STAT1 phosphorylation in STAT1+/+ and STAT1-/- human fibrosarcoma cell lines treated or not (NT) for 1h with 10 uM MNNG and analyzed at the indicated time points. α-actin was used as loading control. 100 ng/ml γ-IFN treatment for 30 min was used as positive control. From [1].
Antibody information

Rabbit monoclonal IgG

Company: Cell Signaling

Antibody: STAT1

Catalog number: 9167

Summary: Rabbit monoclonal IgG against a synthetic phosphopeptide corresponding to residues surrounding Tyr701 of human Stat1. Reacts with human and mouse. Suitable for western blot, immunoprecipitation, immunohistochemistry (paraffin), immunofluorescence/immunocytochemistry, flow cytometry and ChIP.

Validation Method

Western blot

Sample

STAT1+/+ and STAT1-/- human fibrosarcoma cells. Cells were lysed in NRSB minus buffer (62.5 mM Tris pH 6.8, SDS 2%, glycerol 10%).

Primary incubation

1/1,000 dilution.

Detection

Chemiluminescence reagent (Amersham, Orsay, France) using a Chemidoc apparatus and the Image Lab 4.2 software (Bio‐Rad, Marnes‐la‐Coquette, France).

Disclaimer

If the antibody described in this summary is a polyclonal antibody, since polyclonal antibodies are of limited quantity, please inquire the supplier whether any current polyclonal antibody with the same catalog number is exactly the same as the one described in this summary. Sometimes, different bleeds or different animals are used, usually with a different lot number. In such cases, the result in this summary may not apply to the new antibody with the same catalog number.
