SPACR antibody | knockout validation | Santa Cruz sc-377366

This is a knockout-validated antibody summary, based on the publication "Proteoglycan IMPG2 Shapes the Interphotoreceptor Matrix and Modulates Vision", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Antibody information

Mouse monoclonal IgG1 κ

Company: Santa Cruz

Antibody: SPACR

Catalog number: sc-377366

Summary: Mouse monoclonal antibody against amino acids 121-420 mapping within an internal region of mouse SPACR (IMPG1). Recommended for detection of SPACR of mouse, rat and human origin by western blot, immunoprecipitation, immunofluorescence and ELISA.

Validation Method

Immunohistochemistry

Sample

WT and IMPG1-KO mouse retina cryosections.

Blocking agent

PBS with 5% goat sera, 0.5% Triton X-100, 0.05% sodium azide) at room temperature for 1 h.

Primary incubation

1:2,000 dilution overnight at 4°C.

Secondary incubation

For 2 h at room temperature.

Detection

Images were acquired and processed with a Nikon C2 laser-scanning

confocal microscope using excitation wavelengths of 405, 488, 543, and

647 nm.

Figure Immunohistochemistry of the proteoglycans IMPG1, IMPG2, and PNA lectin from wild-type (left), IMPG1 (middle), and IMPG2 KO (right) mice. IMPG1 KO mice show the absence of IMPG1 (green) and preserved IMPG2 (red) staining at the IS. Please see Figure 2B,C in the article [1].
