Western blot
Control and SNX27-KO HeLa cells. Cells were homogenized in buffer containing 20 mM
HEPES, pH 7.4, 250 mM sucrose, 1 mM EDTA, 10 mM Na4P2O7, 30 mM NaF,
2 mM Na3VO4, 0.5 mM AEBSF and protease-inhibitor cocktail with a 22 3/4-gauge
needle.
5% BSA or 5% skim milk in PBS containing 0.1% Tween-20 for 1 h at room temperature.
1:1,000 dilution overnight at 4 °C.
1:4,000 dilution HRP-conjugated goat anti-mouse secondary antibody(62-6520, Life Technologies).
Chemiluminescence signals were detected with a Super Signal ECL detection kit (Thermo Scientific).
Immunoblot analysis of whole cell lysates from control HeLa cells (stably expressing Cas9) and SNX27-KO cells. Na+/K+ ATPase is a control plasma-membrane protein; β-tubulin is a loading control. Please see Figure 7b in the article [1].
