Western blot | Immunohistochemistry

Sample

WB: WT, Smurf1−/− MEFs and mouse tissues. Cell lysates were prepared in RIPA buffer with protease inhibitor cocktail. IHC: kidney and colon tissues of WT or Smurf1−/− mice.

Primary incubation

WB: 1:1,000 dilution.

IHC: 1:100 dilution overnight at 4°C

Secondary incubation

IHC: secondary antibodies (Dako) for 30 min.

Detection

WB: Super Signal chemiluminescence kit (Thermo Fisher Scientific).

IHC: BX51 microscope (Olympus), and images were photographed with a 40× objective.
