Sirt5 WT and KO mouse hearts. Tissues were lysed in lysis buffer (50 mM Tris⋅HCl, pH 8.0, 150 mM NaCl, 1 mM EDTA, 1% Nonidet P-40, 5 mM nicotinamide, and protease inhibitor mixture).

Blocking agent

5% BSA or milk in TBST (25 mM Tris⋅HCl, pH 7.4, 150 mM NaCl, and 0.1% Tween-20).

Primary incubation

Diluted in 5% BSA or milk in TBST.

Detection

ECL Plus Western blotting detection reagents (GE Healthcare). Chemiluminescence was recorded by a Storm 860 Imager (Amersham Biosciences).
