Control and Sgk1-3-TKO primary hepatocytes. Cell lysates were prepared in RIPA buffer (50 mM Tris-HCl pH 7.4,150 mM NaCl, 1% Triton x-100, 1% Sodium deoxycholate, 0.1% SDS,1 mM EDTA with proteinase inhibitor cocktail from Roche).

Primary incubation

According to manufacturers’ protocols.

Secondary incubation

1:5,000 dilution goat-anti-rabbit-HRP conjugate (GE Healthcare) in 5% BSA dissolved in TBST.

Detection

West-Pico chemiluminescence substrate (Thermo Pierce).
