Cells were lysed in 1 ml of cold lysis buffer containing 50 mM Tris-HCl, pH 7.6, 150 mM NaCl, 0.1% Triton X-100, 0.75% Brij96, 1 mM sodium orthovanadate, 1 mM sodium fluoride, 1 mM sodium pyrophosphate, 10 mg ml−1 aprotinin, 10 mg ml−1 leupeptin, 2 mM PMSF and 1 mM EDTA for 20 min on ice.

Primary incubation

1:1,000 dilution overnight at 4 °C.

Secondary incubation

1:10,000 dilution horseradish peroxidase-linked anti-rabbit IgG (Jackson ImmunoResearch, #711-035-152) for 1 h at room temperature.

Detection

Chemiluminescence ECL detection kit (Biological industries).
