Immunocytochemistry

Control and Rab18-KO 3T3-L1 preadipocytes. Cells were fixed with 4% paraformaldehyde for 20 min.

10% goat serum in PBS supplemented with 0.1% saponin for 1 h.

1:50–1:200 diluted in PBS supplemented with 0.1% saponin overnight at 4°C.

1:500 diluted Alexa Fluor 488–conjugated donkey anti–mouse IgG (Life Technologies) for 1 h at room temperature.

Images were collected on a Nikon A1R+ laser scanning confocal microscope with CFI Plan Apo 100× oil immersion objective (NA 1.45) and 405/488/561/640 laser.
