Validation Method

Western blot

Sample

Lysates of wild-type and REDD1 -/- mouse embryonic fibroblasts.

Blocking agent

TBS (20mM Tris-base, 150mM NaCl, 50mM KCl) containing 5% milk powder and 0.2% Tween 20.

Primary incubation

Anti-REDD1 antibody diluted 1:800 in TBS containing 1% milk powder and 0.2% Tween 20. Overnight incubation.

Secondary incubation

HRP-conjugated Affinity Pure goat anti-rabbit IgG (Jaclson ImmunoResearch Laboratories) diluted 1:7500 in blocking buffer.

Detection

Clarity Western ECL Blotting reagent (BioRad).
