Sample

Rab7+/+ and rab7−/− MEFs. WB: cells were lysed in the plate on ice with lysis buffer (2% Triton X-100, 80 mM HEPES, 0.4 mM EDTA, pH 6.6) containing complete protease inhibitor cocktail (Roche), 50 mM sodium fluoride, 100 nM sodium orthovanadate, and 50 mM β-glycerophosphate.

Blocking agent

ICC: PBS containing 0.3% saponin, 10% FBS and 0.05% NaN3.

Primary incubation

ICC: 1:100 dilution in blocking buffer overnight at 4°C.

Secondary incubation

WB: secondary goat anti-rabbit (LI-COR, 92632211).

ICC: 1:100 dilution secondary anti-rabbit Alexa488 in blocking buffer for 2 h at RT.

Detection

EB: blots were scanned with an Odyssey imager (LI-COR Biosciences).

ICC: Nikon Eclipse TE2000 fluorescence microscope equipped with a CoolSNAP CCD camera (Photometrics).
