This is a knockout-validated antibody summary, based on the publication "Sox7, Sox17, and Sox18 Cooperatively Regulate Vascular Development in the Mouse Retina", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Company

R and D Systems

Antibody

Polyclonal goat IgG against recombinant human Sox-7

Catalogue No: AF2766

Summary

This antibody was validated in Sox7 conditional knockout mice (Sox7CKO-/-) by Immunostaining [1]. The retinas of wild type and knockout mice were processed and immunostained. The results confirmed the specificity of this antibody as the immunostaining signal was absent in the retinas of knockout mice.

Experimental design
Technique

IHC, immunohistochemistry.

Primary antibody incubation

Primary antibody was diluted in PBSTC (PBS + 0.3% Triton + 0.1mM CaCl2) + 10% normal goat/donkey serum and sections were incubated for 1–7 days at 4°C.

Secondary antibody incubation

Secondary antibodies were diluted in PBSTC + 10% normal goat/donkey serum and sections were incubated for 1–3 days at 4°C.

Detection System

Images were captured with Zeiss LSM700 confocal microscope and processing was done with ImageJ, Photoshop, and Illustrator software.

Results

Sox 7 expression was not detected in Sox7 conditional knockout mice (Sox 7CKO-/-) (see figure 1).
