Validation Method

Western blot | Immunocytochemistry

Sample

WT and PDHA1 KO KYSE450 cells.

Blocking agent

WB: 5% milk (1 hr, room temperature).

ICC: 3% H2O2 (25 mins) and 3% BSA (30 mins).

Primary incubation

WB: overnight at 4°C.

ICC: 1:100 dilution at 4°C, overnight.

Secondary incubation

WB: horseradish peroxidase conjugated (1 hr, room temperature).

Detection

WB: photographic developer (Western Dura Extended Duration Substrate, SuperSignal, USA) and X-ray film.
