Figure 1. From supplemental Figure 3 in [1]. Staining of paraformaldehyde-fixed MEFs from wild-type and Lmna knockout mice using anti-prelamin A antibody.
Antibody information

Rat monoclonal

Company: Millipore

Antibody: Prelamin-A

Catalog number: MABT345 (Clone 7G11)

Summary: Rat monoclonal antibody raised against a peptide corresponding to the C-terminal propeptide of prelamin-A. Supplier recommended for WB and IF. Reacts with mouse prelamin-A.

This antibody does not react with mature prelamin-A lacking the C-terminal propeptide.

Immunocytochemistry

Sample

Wild-type and Lmna -/- mouse embryonic fibroblasts fixed with 4% paraformaldehyde and permeabilised with 0.2% Triton X100 in PBS.

Blocking agent

PBS containing 1% BSA and 0.1% Tween 20.

Primary incubation

Anti-prelamin-A (1μg/ml).

Secondary incubation

Alexa flour595-conjugated donkey anti-rat IgG (Invitrogen, A21209) at 1:200 dilution.

Detection

Delta Vision Elite image recovery system (Applied Precision Inc.).

Disclaimer

If the antibody described in this summary is a polyclonal antibody, since polyclonal antibodies are of limited quantity, please inquire the supplier whether any current polyclonal antibody with the same catalog number is exactly the same as the one described in this summary. Sometimes, different bleeds or different animals are used, usually with a different lot number. In such cases, the result in this summary may not apply to the new antibody with the same catalog number.
