Western blot

Sample

WT and Prnp-KO N2a, C2C12 and NMuMG mouse cells. Cells were lysed with a buffer consisting of 50 mM Tris, pH 8.0, 150 mM NaCl, 1% NP-40, and Roche Complete Protease Inhibitor Cocktail.

Blocking agent

Skim milk.

Primary incubation

1∶5,000 dilution overnight at 4°C.

Secondary incubation

1∶10,000 dilution anti-mouse secondary antibody (170-6516; Bio-Rad, Mississauga, ON, Canada).

Detection

ECL reagent (RPN2106; GE Healthcare, Baie d'Urfe, QC, Canada). Signals were detected with either X-ray film or the Li-COR Odyssey Fc digital imaging system (NE, USA).
