Validation Method

Western blot; IHC

Sample

Kidneys from Prdx6+/+ and Prdx6-/- mice. WB: Tissues were homogenized in 500 μl of cold homogenization buffer (10 mM Tris pH 7.4, 150 mM NaCl, 1 mM EGTA, 1 mM EDTA, 2 mM PMSF, and protease inhibitors). IHC: Mouse kidneys were fixed in 10% neutral buffered formalin, and paraffin-embedded.

Primary incubation

IHC: 1:500 dilution overnight at 4°C.

Secondary incubation

IHC: 1:200 dilution fluorochrome conjugated secondary antibodies for 1hr at room temperature.

Detection

IHC: Zeiss LSM880 confocal microscope.

Kidney sections from Prdx6+/+ and Prdx6-/- mice were used to test the specificity of the anti PRDX6 antibodies Ab59543 (green) and Ab16947 (red) (A) Scale bars: 20 µm. Western blots with three samples each of Prdx6+/+ and Prdx6-/- total kidney lysate with a human kidney total (HKT) lysate as a control were probed with Ab59543 (B) and Ab16947 (C). Actin and tubulin were used as loading controls respectively. The sample preparations were carried out under reducing conditions. Please see Supplementary Figure 1 in the article [1].
