This is a knockout-validated antibody summary, based on the publication "PARK2/Parkin-mediated mitochondrial clearance contributes to proteasome activation during slow-twitch muscle atrophy via NFE2L1 nuclear translocation", as cited below [1]. This antibody was also validated for Western blot in the article "Characterization of Dopaminergic System in the Striatum of Young Adult Park2-/- Knockout Rats" (see figure 1a) [2] and in the article "Parkin Promotes Mitophagic Cell Death in Adult Hippocampal Neural Stem Cells Following Insulin Withdrawal" (see figure 6a) [3] and "Pathogenic Pathways in Early-Onset Autosomal Recessive Parkinson's Disease Discovered Using Isogenic Human Dopaminergic Neurons" for western blot knockout validation (see figure 2h, s3g) [4]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).
Mouse monoclonal IgG2b
Company: Cell Signaling Technology
Antibody: Parkin
Catalog number: 4211S
Summary: Mouse monoclonal IgG2b against an epitope mapping to the carboxy terminus of human recombinant parkin. Reacts with human, mouse and rat. Suitable for western blot and immunoprecipitation.
Western blot | Immunohistochemistry
Soleus muscles from control and Parkin-KO mice. WB: muscles were homogenized in 10 volumes of 50 mM TRIS-HCl (pH 7.4) containing 0.15 M NaCl, 1 mM EDTA, 1% Triton X-100, 0.5% sodium deoxycholate, 0.1% SDS, a protease inhibitor cocktail (Roche Diagnostics, 11836170001), and a phosphatase inhibitor cocktail (Roche Diagnostics, 04906837001). IHC: mouse hind limbs were cryosectioned (10 µm thick).
IHC: Alexa 594-conjugated secondary antibodies (A11032, Molecular Probes).
Western blot analysis of Parkin in soleus muscles from control and Parkin-KO mice at 7 d after denervation. GAPDH was used as loading control. Immunofluorescent micrographs of denervated (7 d) or innervated soleus muscles of the indicated genotypes stained with anti-PARK2 (red) or anti-TOMM20 (mitochondrial marker, green) antibodies and Hoechst 33342 (nucleus, blue). Please see Figure 3a,d in the article [1].
IHC is not mentioned in the antibody datasheet from the supplier CST.
