This is a knockout-validated antibody summary, based on the publication "GPR37 regulates macrophage phagocytosis and resolution of inflammatory pain", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).
Mouse monoclonal IgG1 (kappa light chain)
Company: Santa Cruz
Antibody: Pael-R
Catalog number: sc-390110
Summary: Mouse monoclonal antibody against amino acids 31-280 mapping within an N-terminal extracellular domain of human Pael-R (GPR37). Recommended for detection of Pael-R of mouse, rat and human origin by western blot, immunoprecipitation, immunofluorescence and ELISA.
Western blot | Immunohistochemistry
WB: Brain, spinal cord (SC), hind paw skin, and pMΦs from WT and Gpr37–/– mice.
IHC: Hind paw dermis of WT and Gpr37–/– mice. After the 4% PFA perfusion, hind paw glabrous skins were removed and postfixed in the same fixative overnight.
IHC: 2% goat or donkey serum for 1 hour at room temperature.
WB: 1:2,000 dilution.
IHC: 1:500 dilution overnight at 4°C.
WB: HRP-conjugated secondary antibody.
IHC: 1:400 dilution cyanine 3– (Cy3-) or FITC-conjugated secondary antibodies (Jackson ImmunoResearch) for 2 hours at room temperature.
WB: ECL solution (Pierce, Thermo Fisher Scientific) and Bio-Rad ChemiDoc XRS for 1 to 10 minutes.
IHC: Nikon fluorescence microscope, and images were captured with a CCD SPOT camera (SPOT Imaging).
IHC showing the colocalization of GRP37 and CD68 in the hind paw dermis of WT (A) but not Gpr37–/– (B) mice. Boxes in A indicate an enlarged cell. Blue DAPI staining labels all nuclei in the skin. Scale bar: 100 μm; 45 μm (original scale in inset). Western blot showing a single band of GRP37 in lysates of brain, spinal cord (SC), hind paw skin, and pMΦs. Note that the band is absent in Gpr37–/– mice. Please see Figure 1b and 1g in the article [1].
