PSD-95 antibody | knockout validation | NeuroMab 75-028

This is a knockout-validated antibody summary, based on the publication "PSD-95 stabilizes NMDA receptors by inducing the degradation of STEP61", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Antibody information

Mouse monoclonal IgG2a

Company: NeuroMab

Antibody: PSD-95

Catalog number: 75-028

Summary: Mouse monoclonal IgG2a against a fusion protein corresponding to amino acids 77-299 (PDZ domains 1 and 2) of human PSD-95. Reacts with human, rat, and mouse. Suitable for immunoblot, immunocytochemistry,

immunogold EM and immunoprecipitation.

Validation Method

Western blot

Sample

Brain tissues from WT and PSD-95 KO mice. Tissues were homogenized with ice-cold TEVP buffer: 320 mM sucrose, 10 mM Tris⋅HCl (pH 7.5), 5 mM EDTA, 1× protease inhibitor mixture (Roche, 11697498001), and phosphatase inhibitor mixture II (Sigma, catalog no. P5726) and III (Sigma, catalog no. P0044) and subsequently fractionated.

Figure

WT or PSD-95–KO adult mouse forebrain was subjected to fractionation, and the PSD fraction was isolated and immunoblotted with the indicated antibodies. Please see Figure 6a in the article [1].

Clone note

The same clone (K28/43) is sold as BioLegend 810401; Neuromab 73-028, 75-028; EMD Millipore MABN68.
