Validation Method

Western blot | Immunohistochemistry

Sample

Hypothalamus of of WT and RIIβ KO mice. WB: The tissue was homogenized in 200 μl lysis buffer (50 mM Tris-Cl, pH7.4, 150 mM NaCl, 5 mM EDTA, 1% Triton X-100, 0.5% deoxycholic acid) supplemented with protease and phosphatase inhibitors (5 mM NaF, 1 mM Na3VO4, PhosStop (Roche), cOmplete Ultra protease inhibitors (Roche)). IHC: Brains were removed and postfixed, followed by cryopreservation in 30% sucrose solution (w/v) overnight and subsequent freezing in OCT compound (Tissue-Tek).
