This is a knockout-validated antibody summary, based on the publication "Epidermal or dermal specific knockout of PHD-2 enhances wound healing and minimizes ischemic injury", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Figure 1. Immunofluorescent staining of PHD-2 on healed wounded skin and adjacent normal unwounded skin of wild type, heterozygous K14-Cre/homozygous floxed PHD-2, and heterozygous Col1α2-Cre-ER/homozygous floxed PHD-2 mice at 400× magnification. From [1].
Antibody information

Rabbit polyclonal

Company: Novus

Antibody: PHD-2

Catalog number: NB100-2219

Summary: Rabbit polyclonal against a synthetic peptide made to an internal portion of mouse PHD2/HIF Prolyl Hydroxylase 2 (between residues 300-400). Reacts with human, mouse and rat. Recommended for western blot, immunoprecipitation, immunocytochemistry, and immunohistochemistry (paraffin).

Validation Method

Immunohistochemistry

Sample

Skin of wild type, heterozygous K14-Cre/homozygous floxed PHD-2, and heterozygous Col1α2-Cre-ER/homozygous floxed PHD-2 mice. Tissues were fixed in 4% paraformaldehyde and embedded in paraffin for sectioning.

Primary incubation

1∶100 dilution.

Secondary incubation

1∶200 dilution Alexa Fluor 594 Goat Anti-Rabbit IgG (Invitrogen) at room temperature for 1 hour.

Detection

A Zeiss Axioplan 2 fluorescence microscope was used to image the slides (Carl Zeiss, Thornwood, New York).

Disclaimer

If the antibody described in this summary is a polyclonal antibody, since polyclonal antibodies are of limited quantity, please inquire the supplier whether any current polyclonal antibody with the same catalog number is exactly the same as the one described in this summary. Sometimes, different bleeds or different animals are used, usually with a different lot number. In such cases, the result in this summary may not apply to the new antibody with the same catalog number.
