Western blot

Sample

WT and PEPT1-KO iPSC--derived intestinal epithelial-like cells (IECs). The cells were homogenized with RIPA Lysis and Extraction Buffer (Thermo Fisher Scientific) containing a protease inhibitor mixture (Thermo Fisher Scientific).

Blocking agent

1% skim milk in TBS containing 0.1% Tween 20 at room temperature for 1 h.

Primary incubation

At 4°C overnight.

Secondary incubation

Horseradish peroxidase (HRP)-linked secondary antibodies (Cell Signaling Technology) at room temperature for 1 h.

Detection

Chemi-Lumi One Super (Nakalai Tesque) and the signals were read using an LAS-3000 imaging system (Fujifilm).
