Sample

Control and PCM1 KO hTERT RPE-1 cells.

Blocking agent

IC: 3% BSA-0.3% PBST (Triton X-100) for 20 min.

Primary incubation

IC: diluted in 0.1% PBS with Triton X-100 with 3% BSA for 2 hrs at RT.

Secondary incubation

IC: diluted in 0.1% PBS with Triton X-100 with 3% BSA for 2 hrs at RT.

Detection

IC: 63× or 100×, NA 1.4; Axiovert 200M, Carl Zeiss, Germany) equipped with a cooled CCD (Retiga 2000R; QImaging, Surrey, Canada) and MetaMorph.
